      为探讨破壁方法对雨生红球藻中虾青素提取的影响,本研究以雨生红球藻湿藻细胞为原料,比较研究了研磨、酸解、碱解、热蒸汽和酶解等多种破壁处理方法对其细胞总类胡萝卜素和总脂肪提取效率及其组成的影响。结果表明,酸解和酶解处理能够显著提高类胡萝卜素的提取效率,而不影响其组成。而碱解处理导致酯化虾青素的水解,游离虾青素含量比未处理的对照组提高了20倍,同时细胞内的甘油三酯也发生明显降解。热蒸汽和研磨处理对于虾青素和油脂的提取率及其组成影响不明显。因此,酶解和酸解是提高虾青素提取效率较有效预处理方法,并能很好保持虾青素和脂肪酸的结构稳定性和生物活性。

      The effect of cell-wall disruption methods on astaxanthin extraction from wet algal biomass of Haematococcus pluvialis was investigated. The contents and compositions of total carotenoids and total lipid were compared, after various treatments of cell-wall disruption, included grinding, enzymatic hydrolysis, acid hydrolysis, alkaline hydrolysis and hot steam. The results showed that the carotenoids extraction efficiencies were significantly increased, but their compositions were untouched, after either acid hydrolysis or enzymatic hydrolysis. The alkaline hydrolysis caused the degradation of esterified astaxanthin and increased the content of free astaxanthin to 20-folds of the control. Meanwhile, the intracellular triglyceride also significantly degraded. Hot steam and grinding had no obvious effect on the contents and compositions of the astaxanthin and lipid. In conclusion, enzymatic hydrolysis and acid hydrolysis, which maintained the stability and biological activity of the astaxanthin and the fatty acid, were the most effective pretreatment methods for cyst cells of H. pluvialis to improve the extraction efficiency of the astaxanthin.
