四君子汤复方多糖及其单方多糖的免疫调节作用

Immunomodulatory Effects of Composite and Single Polysaccharides from Sijunzi Decoction

    摘要:

    目的:探究四君子汤总多糖的免疫调节作用并解析其可能的作用机制。同时,对比四君子汤总多糖与组方中单味原料多糖的免疫调节作用差异,以期从食品科学与营养学角度阐释经典古方中功能物质的贡献特点,挖掘经典古方中的关键活性物质,为传统方剂功能特性的科学阐释及其在功能食品领域的应用提供理论支撑。方法:采用环磷酰胺构建免疫低下小鼠模型,从体质量变化、免疫器官指数、组织病理学、免疫球蛋白含量及细胞因子分泌等方面,系统性评价四君子汤总多糖及人参、茯苓、炙甘草、白术多糖的免疫调节作用。结果:四君子汤总多糖能显著缓解免疫抑制小鼠体质量下降,恢复脾脏指数和胸腺指数,并提升血清IgA、IgM、IgG及脾脏TNF-α水平,效果优于单一多糖组分;且其还可通过调控α-亚麻酸代谢、花生四烯酸代谢等代谢通路改善代谢紊乱。在单一多糖中,人参多糖效果最为显著,印证其“君药”地位。结论:四君子汤总多糖是四君子汤发挥免疫调节功效的主要活性成分,其免疫调节机制与调节关键代谢通路(如α-亚麻酸代谢)及增强体液免疫和细胞免疫功能密切相关,并且人参多糖是四君子汤总多糖发挥免疫调节功效的主要贡献成分。本研究为食药同源多糖资源的挖掘利用及其作用机制探究提供了新视角,未来需进一步结合转录组学与蛋白组学解析并验证其分子靶点。

    Abstract:

    Objective: This study aimed to investigate the immunomodulatory effects of total polysaccharides from Sijunzi Decoction and elucidate their potential mechanisms, while comparing the differences in immunomodulatory activity between Sijunzi Decoction polysaccharides and polysaccharides from its individual constituents (Panax ginseng, Poria cocos, Atractylodes macrocephala Koidz, Glycyrrhiza uralensis Fisch). The research sought to interpret the contribution characteristics of functional components in classical formulas from the perspective of food science and nutrition, explore key bioactive substances in classical formulas, and provide theoretical support for the scientific interpretation of traditional prescriptions' functional characteristics and their applications in functional foods. Methods: An immunosuppressed mouse model was established using cyclophosphamide. The immunomodulatory effects of Sijunzi Decoction polysaccharides and individual polysaccharides were systematically evaluated based on body weight changes, immune organ indices, histopathology, and levels of immunoglobulins (IgA, IgM, IgG) and cytokines. Results: Sijunzi Decoction polysaccharides significantly alleviated weight loss in immunosuppressed mice, restored spleen and thymus indices, and enhanced serum IgA, IgM, IgG levels and TNF-α secretion, with superior effects compared to individual polysaccharides. Furthermore, Sijunzi Decoction polysaccharides improved metabolic disorders by regulating key pathways such as α-linolenic acid metabolism and arachidonic acid metabolism. Among individual polysaccharides, ginseng polysaccharides exhibited the most pronounced effects, affirming their role as the 'monarch drug' in the formula. Conclusion: Sijunzi Decoction polysaccharides serves as the primary active component responsible for the immunomodulatory effects of Sijunzi Decoction, mediated through the regulation of critical metabolic pathways (e.g., α-linolenic acid metabolism) and enhancement of both humoral and cellular immune functions. Ginseng polysaccharides were identified as the major contributing component within Sijunzi Decoction polysaccharides. This study offers novel insights into the utilization of medicinal-edible homologous polysaccharides and their mechanisms, though future research integrating transcriptomics and proteomics is required to validate molecular targets.
