    以多糖为代表的复杂碳水化合物结构是其发挥生物学功能的基础,而由于多糖结构本身的复杂性,加之缺乏适合其结构特点的理论指导,研究进展较为缓慢。针对上述问题,本文旨在全面介绍多糖从提取、分离纯化到结构解析的关键方法,着重阐述构建多糖结构解析方法与理论体系的关键要素:通常情况下多糖具有“相对有序性”的结构特征;其次,多糖分离纯化可以建立在“三基于、三阶段”的“全流程”纯化策略上,对于纯化获得的均一组分应采用“多维性”的方法和手段予以综合评价纯度;最后,采用以“部分降解-甲基化-核磁共振(NMR)”为核心技术的方法对多糖结构进行科学、精确地解析,并结合“多糖结构的相对有序性”理论,以期获得可重复的结果。最后,本文概述了多糖结构研究领域未来发展的方向。

    Abstract:

    The biological function of complex carbohydrates represented by polysaccharide is fundamentally determined by their structural composition. However, the complexity inherent in polysaccharides structure and the lack of adequate theoretical guidance for their characterization has impeded progress in this field. To address these issues, this review aimed to comprehensively introduce key methods for polysaccharide extraction, isolation, purification, and structural analysis, with a particular focus on the essential components required for constructing analytical methods and theoretical system for polysaccharide structure characterization. Firstly, polysaccharides generally exhibit the structural characteristic of 'relative ordered structure'. Subsequently, effective separation and purification of polysaccharides can be achieved through the implementation of 'three consecutive and progressive stages' purification strategy, which involves a universal evaluation of the homogeneous components obtained from purification using a multidimensional approach. Additionally, to ensure scientific precision and repeatability in polysaccharide structural characterization, the core technology of 'partial degradation-methylation-NMR' is employed. An assessment was also conducted to determine adherence to the theory of 'relative ordered structure' to obtain replicable outcomes. Finally, a comprehensive overview of the future research directions in polysaccharide structure analysis was summarized.
