    本文探讨姬松茸提取物的降血糖作用及对糖尿病大鼠肠道菌群的影响。对链脲佐菌素诱导的糖尿病大鼠灌胃姬松茸乙醇提取物和姬松茸乙酸乙酯提取物,于4周后采集大鼠血液、肝脏和盲肠内容物,测定血糖水平并用Ⅱlumina HiSeq平台进行高通量测序。结果表明:给予姬松茸提取物后,糖尿病大鼠毛发变得更加润泽,精神状态有所改善,多饮、多尿、多食且消瘦的症状有所减轻。姬松茸提取物能够降低糖尿病大鼠的空腹血糖值,其中姬松茸乙酸乙酯提取能够显著降低空腹血糖值(P<0.05),改善糖尿病大鼠肝脏细胞肿胀,减少厚壁菌门丰度,增加拟杆菌门丰度。可见,姬松茸提取物可通过调节糖尿病大鼠肠道菌群的丰度,增加肠道菌的多样性来恢复肠菌群的结构,从而发挥治疗糖尿病的作用。

    Abstract:

    This study was designed to investigate the hypoglycemic effect of Agaricus blazei Murrill extracts and its beneficial effects on gut microbiota in diabetic rats. The diabetic rats were treated with ethanol and ethyl acetate extract of Agaricus blazei Murrill for four weeks. The samples of blood, liver tissues and feces were collected to evaluate the level of fasting blood glucose and Ⅱlumina HiSeq platform was used for high throughput sequencing. Results showed that supplementation with Agaricus blazei Murrill extracts could decrease the urine output and improve the mental state in diabetic rats. The symptoms of polydipsia, polyuria, polyphagia, and weight loss in diabetic rats were alleviated. Agaricus blazei Murrill extracts could cause reduction of the level of fasting blood glucose. Specifically, the ethyl acetate extract could significantly decrease the level of fasting blood glucose in diabetic rats (P<0.05). The Agaricus blazei Murrill treated rats caused the up-regulation of Bacteroidetes and down-regulation of Firmicutes. The abundance and diversity of gut microbiota in diabetic rats were improved. The study suggested that Agaricus blazei Murrill extracts may play a role in the treatment of diabetes through regulating the structure of gut microbial.
