    本实验利用N-硝基-L-精氨酸甲酯(L-NAME)诱导的高血压小鼠模型,探究制备的黑豆血管紧张素转化酶(ACE)抑制肽(BSP,<3 ku)对高血压小鼠的预防、改善功效及机理。将小鼠分为正常组、高血压模型组、赖诺普利药物组、BSP低、中、高剂量组(25,50,100 mg/kg)、L-NAME诱导同时灌胃BSP高剂量预防组(L-NAME+BSP组),连续灌胃4周后,进行血浆生理指标检测及心脏、肾脏组织病理切片观察。结果表明:BSP中、高剂量组对高血压小鼠具有显著的降压作用(P<0.05),能够抑制小鼠血浆中ACE、血管紧张素Ⅱ(AngⅡ)、单核细胞趋化蛋白-1(MCP-1)、白细胞介素-6(IL-6)及内皮缩血管肽-1(ET-1)含量的上升(P<0.05),显著升高一氧化氮(NO)含量(P<0.05),避免小鼠肾素-血管紧张素(RAS)系统紊乱,提示BSP具有抗炎及维持正常血管细胞功能的功效,减轻了小鼠心脏损伤、纤维化和肾脏炎症浸润程度。同时,L-NAME延缓了小鼠高血压症状的发生速度,显示出良好的预防作用。结论:该食源性黑豆ACE抑制肽通过调节RAS系统起到预防及改善小鼠高血压症的作用。

    Abstract:

    Black soya ACE inhibitory peptide (<3 ku) was prepared, and a hypertensive mouse model was established by using N-nitro-L-arginine methyl ester induction, the mice were divided into control group, hypertensive model group, lisinopril drug group, the BSP low, medium and high dose groups (25, 50, 100 mg/kg) and L-NAME-induced simultaneous gavage BSP high-dose prophylaxis group (L-NAME+BSP). After 4 weeks of continuous gavage, plasma physiological indices were tested and histopathological sections of heart and kidney were observed to investigate the preventive and ameliorative efficacy of BSP and its mechanism of action on hypertensive mice. The middle and high dose groups of BSP had significant antihypertensive effects on hypertensive mice (P<0.05), inhibited the rise of ACE, AngⅡ, MCP-1, IL-6 and ET-1 in plasma of mice (P<0.05), and significantly increased NO content(P<0.05), avoided the disorders of the RAS system of mice, and possessed the efficacy of anti-inflammation and maintenance of the normal function of the vascular cells, and reduced the degree of cardiac damage, fibrosis and renal inflammatory infiltration in mice. Meanwhile, the L-NAME+BSP group delayed the onset of hypertension symptoms in mice, showing a favorable preventive effect. This food-derived black soya ACE inhibitory peptide acts to prevent and ameliorate hypertension in mice by modulating the RAS system.
