    摘要:

    目的:对金枪鱼蛋白水解产物中的免疫活性组分进行分离、鉴定。方法:通过胰蛋白酶水解金枪鱼蛋白,采用葡聚糖凝胶色谱和液相色谱-质谱联用(LC-MS/MS)方法进行分离、纯化和鉴定。以小鼠巨噬细胞RAW264.7的相对增殖率、吞噬能力、一氧化氮(NO)生成能力、细胞周期分布为评价指标,筛选具有较强免疫活性的组分,并鉴定组分结构。结果:经SephadexG-25和SephadexG-15层析分离、纯化,得到的FF1组分具有较强的免疫活性,其RAW264.7细胞相对增殖率为73.66%,吞噬能力为70.04%,NO生成能力为4.82 μmol/L。结论:经SephadexG-15分离得到的FF1组分具有较强的免疫活性,并从FF1组分中筛选出2条潜在的免疫活性肽序列,分别为Gly-Asp-Ala-Glu-Met-Glu-Ala-Tyr-Gly-Lys(GDAEMEAYGK)和Leu-Leu-Tyr-Gly-Gly-Ser-Val-Thr-Gln-Thr-Cys-Lys(LLYGGSVTQTCK)。研究结果可为金枪鱼免疫活性肽的开发、利用提供重要的理论参考。

    Abstract:

    Purpose: To screen and identify immunologically active components in the hydrolysate of tuna protein. Method: Tuna protein was hydrolyzed using trypsin, and separation, purification, and identification were performed using a combination of Sephadex G-25 gel chromatography and liquid chromatography-mass spectrometry (LC-MS/MS). The evaluation criteria included the relative proliferation rate, phagocytic ability, nitric oxide(NO) production capacity, and cell cycle distribution of RAW264.7 cells. Components with strong immunological activity were selected, and their structures were identified. Results: After purification by Sephadex G-25 and Sephadex G-15 chromatography, the isolated FF1 fraction exhibited strong immunological activity, with a relative proliferation rate of 73.66%, phagocytic ability of 70.04%, and NO production capacity of 4.82 μmol/L in RAW264.7 cells. Conclusion: The FF1 fraction obtained after Sephadex G-15 separation demonstrated strong immunological activity, and two potential immunologically active peptide sequences were screened from this fraction, namely, Gly-Asp-Ala-Glu-Met-Glu-Ala-Tyr-Gly-Lys (GDAEMEAYGK) and Leu-Leu-Tyr-Gly-Gly-Ser-Val-Thr-Gln-Thr-Cys-Lys (LLYGGSVTQTCK). This study provides important theoretical references for the development and utilization of immunologically active peptides from tuna.
