    摘要:

    细胞培养肉技术作为通过培养动物细胞获取可食用肉类的新兴技术,近十年虽取得显著研究进展,但仍面临成本高昂,生产规模有限,产品品质低下等问题,制约了其进一步发展。本文综述细胞培养肉的生物工程学基础与最新研究进展,并针对关键技术挑战进行分析,提出如下潜在突破途径与未来研究方向:1)简化与标准化分离流程、优化分选策略提高种子细胞获取效率,加速高性能优质细胞株开发,降低细胞培养肉生产成本。2)基于白蛋白、生长因子等成分开发无血清培养基,利用生物合成手段降低关键成分的生产成本,实现低成本无血清培养基研发。3)依据种子细胞生长特性筛选适宜生物反应器,悬浮驯化种子细胞突破贴壁依赖特性,建立细胞规模化悬浮培养体系,推进细胞规模化放大生产。4)优选组织构建材料并探究材料复配策略,利用三维结构设计调控细胞成肌分化,实现细胞培养肉产品品质的显著提升。本文针对细胞培养肉低成本、规模化、高品质生产的生物工程学基础与关键技术挑战进行分析,以期为细胞培养肉未来发展提供参考。

    Abstract:

    As a new technology to produce edible meat through animal cell culture, cultured meat has made remarkable progress in the past decade. However, cultured meat still faces problems including high-cost price, limited production scale, and low product quality, which restrict its further development. This paper reviewed the bioengineering basis and the latest research progress of cultured meat, analyzed the key technical challenges, and proposed potential breakthrough approaches and future research prospects: 1) Simplifying and standardizing the separation process and optimizing the sorting strategy to improve the efficiency of seed cell acquisition, and accelerating the development of cell lines with high performance, the production costs of cultured meat can be reduced. 2) The development of serum-free media based on key components such as albumin and growth factors, and the use of biosynthesis methods to reduce the production cost of key components, can achieve the development of low-cost serum-free media. 3) Selecting appropriate bioreactors according to the growth characteristics of seed cells and adapting cells to suspension culture to break through the adhesion dependent properties are conducive to establish scale-up cell culture suspension system and promote scale-up cell expansion production. 4) Optimizing tissue construction materials by exploring materials compounding strategies, and regulating myogenic differentiation through material three-dimensional structure design can achieve significant improvement in the quality of cultured meat products. In this paper, the bioengineering basis and the key technical challenges of low-cost, scale-up and high-quality production of cultured meat have been analyzed and prospected, in order to provide reference for the future development of cultured meat.
