    摘要:

    目的:探究高压辅助水提和酶提苹果皮膳食纤维对小鼠溃疡性结肠炎的预防作用。方法:通过葡聚糖硫酸钠构建结肠炎小鼠模型,采用酶联免疫法、组织病理学分析、16S rRNA高通量测序、气相色谱等方法,分析苹果皮膳食纤维干预后,小鼠肠道菌群和短链脂肪酸的变化及其与肠道炎症和血清氧化应激水平间的关系。结果:苹果皮膳食纤维可显著降低小鼠肠道炎症水平,改善结肠组织结构,上调超氧化物歧化酶,下调丙二醛、髓过氧化物酶表达量,增加肠道短链脂肪酸含量,证明高压辅助水提苹果皮膳食纤维比酶提苹果皮膳食纤维对小鼠结肠炎缓解效果更好。肠道菌群分析结果表明,肠道微生物多样性与苹果皮膳食纤维干预呈剂量依赖性,苹果皮膳食纤维干预后可富集小鼠肠道拟杆菌门、毛螺菌属,降低变形菌门、副萨特氏菌属丰度。结论:苹果皮膳食纤维可降低肠道炎症,调节肠道菌群。本研究为预防肠炎提供更安全有益的选择,并为开发苹果皮膳食纤维功能性食品提供理论依据。

    Abstract:

    Objective: To explore the preventive effect of high pressure assisted water extraction and enzymatic extraction of apple peel dietary fiber on ulcerative colitis in mice. Methods: A mouse model of colitis was constructed by dextran sulfate sodium. Enzyme-linked immunosorbent assay, histopathological analysis, 16S rRNA high-throughput sequencing, gas chromatography and other methods were used to analyze the changes of intestinal flora and short-chain fatty acids in mice after apple peel dietary fiber intervention and their relationship with intestinal inflammation and serum oxidative stress levels. Results: Apple peel dietary fiber could significantly reduce the level of intestinal inflammation in mice, improve the structure of colon tissue, up-regulate superoxide dismutase, down-regulate the expression of malondialdehyde and myeloperoxidase, and increase the content of short-chain fatty acids in the intestine. It was proved that the high-pressure-assisted water-extracted apple peel dietary fiber was better than the enzyme-extracted apple peel dietary fiber in alleviating colitis in mice. The results of intestinal flora analysis showed that the intestinal microbial diversity was dose-dependent with the intervention of apple peel dietary fiber. After the intervention of apple peel dietary fiber, the intestinal Bacteroidota and Lachnospira of mice were enriched, and the abundance of Proteobacteria and Parasutterella was reduced. Conclusion: Apple peel dietary fiber could reduce intestinal inflammation and regulate intestinal flora. This study provided a safer and more beneficial choice for the prevention of enteritis, and provides a theoretical basis for the development of apple peel dietary fiber functional foods.
