桑椹花青素高抗氧化营养片剂的制备及理化性质研究

    摘要:

    为克服花青素浆果类产品不耐保存及产品类型单一的问题,以桑椹花青素为主料,采用湿法制粒技术制备花青素片剂。其最佳的组分和含量分别为填充剂甘露醇-淀粉复合物(质量比为1∶1)、润湿剂体积分数为50%乙醇(0.80 mL/100 g)、润滑剂硬脂酸镁(质量分数为0.5%)及矫味剂柠檬酸-阿斯巴甜(质量比为1∶2)。在单因素实验基础上,采用响应面法优化了片剂制备的相关参数,得到最佳制备工艺为粒径1.18 mm、质量0.1500 g及含水量5.3%,在此条件下制备的花青素片剂感官评价分数最高。体外抗氧化性研究表明,桑椹花青素片剂对DPPH自由基清除率、超氧阴离子自由基清除率及羟基自由基清除率分别达80%,74%和75%,并可有效将Fe3+还原为Fe2+,显示片剂具有较强的抗氧化能力。进一步研究发现,该片剂对金黄色葡萄球菌、大肠杆菌及枯草芽孢杆菌均具有抑制活性,且表现出剂量依赖效应,抑制效果为金黄色葡萄球菌>大肠杆菌>枯草芽孢杆菌。

    Abstract:

    This study employed wet granulation technology to formulate mulberry anthocyanin tablets to address the issues of inadequate storage stability and limited product forms in anthocyanin-rich berry products. The optimal formulation and content were determined as follows: filler (mannitol-starch complex, mass ratio 1∶1), wetting agent (volume fraction 50% ethanol, 0.80 mL/100 g), lubricant (magnesium stearate, mass fraction 0.5%), and flavoring agent (citric acid-aspartame, mass ratio 1∶2). Through single-factor experiments followed by response surface methodology optimization, it was found that the best tablet preparation process resulted in particles with a size of 1.18 mm, weight of 0.1500 g, and moisture content of 5.3%. These conditions led to the highest sensory evaluation for the produced anthocyanin tablets. In vitro antioxidant studies demonstrated that mulberry anthocyanin tablets exhibited an DPPH radical scavenging rate of 80%, superoxide anion radical scavenging rate of 74%, hydroxyl radical scavenging rate of 75%, and effectively reduced Fe3+ to Fe2+, indicating strong antioxidant capacity. Furthermore, these tablets displayed dose-dependent inhibitory activity against Staphylococcus aureus, Escherichia coli, and Bacillus subtilis; among them Staphylococcus aureus showed the highest inhibition followed by Escherichia coli and Bacillus subtilis.
