    摘要:

    目的:探究水解蛋黄粉(EYH)的理化性质及对SD大鼠长骨生长的影响。方法:采用高效凝胶过滤色谱法、高效液相色谱法测定脱脂蛋黄粉(EYP)和EYH的相对分子质量分布和氨基酸组成。用生长期大鼠进行促进长骨生长功能活性的比较,测定其基本生长指标、骨长度和骨指数、骨生物力学、血清中生化指标。结果:相对分子质量分布测定结果表明,EYH中<1 000 u的肽占比之和为88.97%。氨基酸组成测定结果表明,EYH中7种可增加骨密度的氨基酸占比高于EYP。动物实验表明,通过EYP和EYH的干预能促进骨长度和骨指数增加,而EYH对骨长度和骨指数的促进作用均达到显著性水平,分别增加了2.06%(P<0.05)和9.42%(P<0.05),优于EYP。骨生物力学实验结果表明,EYP和EYH均能显著增加股骨抗弯强度、弹性模量。与CON组相比,EYH处理组分别增加了38.56%(P<0.05)和52.14%(P<0.05),EYP组分别增加了32.51%(P<0.05)和42.63%(P<0.05),相比之下,EYH组增加得更多,说明其具有更好的强化骨骼、增加韧性的能力。大鼠血清生化指标测定结果表明,EYH组在血清中骨形成标志物PⅠNP的含量比CON组提高了18.86%(P<0.05),比EYP组提高了13.27%(P<0.05)。TGF-β1是促进骨形成的重要生长因子,与CON相比,EYH组与EYP组血清中TGF-β1含量分别增加了52.25%(P<0.05)和26.68%(P<0.05)。GH/IGF-1轴对于调节生长和代谢具有重要作用,可以促进骨骼生长。与CON组相比,EYH组与EYP组血清中GH含量分别增加了22.32%(P<0.05)和12.64%(P<0.05),同时EYH组血清中IGF-1含量比CON组和EYP组分别增加了5.04%和17.20%(P<0.05)。EYP可以提升血清中TGF-β1和GH的含量,而EYH的作用更为显著,不仅显著提升大鼠血清中TGF-β1、GH和IGF-1的含量,同时还能增加骨形成标志物PⅠNP的含量,降低骨吸收标志物CTX-Ⅰ的含量。

    Abstract:

    Objective: To explore the physical and chemical properties of hydrolyzed egg yolk powder and its effect on the growth of long bone in SD rats, and to clarify the necessity of developing hydrolyzed egg yolk powder. Methods: the relative molecular weight distribution and amino acid composition of EYP and EYH were determined by high performance gel filtration chromatography and high performance liquid chromatography; The growth promoting activity of long bone in growing rats was compared, and the basic growth indexes, bone length and bone index, bone biomechanics and biochemical indexes in serum were measured. Results: the relative molecular weight distribution showed that the total proportion of peptides < 1 000 u in EYH was 88.97%; The determination of amino acid composition showed that the proportion of 7 kinds of amino acids that can increase bone mineral density in EYH was higher than that in EYP; The results of animal experiments showed that the intervention of EYP and EYH could promote the increase of bone length and bone index, and the promotion effect of EYH on bone length and bone index reached a significant level, increased by 2.06% (P<0.05) and 9.42% (P<0.05), respectively, which was better than EYP; The results of bone biomechanical experiments showed that EYP and EYH could significantly increase the bending strength and elastic modulus of femur. Compared with CON group, the EYH treatment group increased by 38.56% (P<0.05) and 52.14% (P<0.05), and the EYP group increased by 32.51% (P<0.05) and 42.63% (P<0.05), respectively. In contrast, the EYH group increased more, indicating that it has better ability to strengthen bones and increase toughness; The determination of biochemical indexes in rat serum showed that the content of PINP in EYH group was 18.86% higher than that in CON group (P<0.05), and 13.27% higher than that in EYP group (P<0.05). TGF-β1 is an important growth factor to promote bone formation. Compared with CON group, the content of TGF-β1 in serum of EYH group and EYP group increased by 52.25% (P<0.05) and 26.68% (P<0.05), respectively. GH/IGF-1 axis plays an important role in regulating growth and metabolism, and can promote bone growth. Compared with CON group, the serum GH content in EYH group and EYP group increased by 22.32% (P<0.05) and 12.64% (P<0.05) respectively, and the serum IGF-1 content in EYH group increased by 5.04% and 17.20% (P<0.05) respectively compared with CON group and EYP group. EYP can increase the contents of TGF-β1 and GH in serum, while EYH has a more significant ability to increase the contents of TGF-β1, GH and IGF-1 in serum of rats, at the same time, it can increase the content of PINP, a marker of bone formation, and reduce the content of CTX-I, a marker of bone resorption.
