玫瑰可溶性膳食纤维的提取及其生理活性和降血糖功能研究

    以玫瑰花渣为材料,采用化学碱法提取玫瑰花膳食纤维(DF),通过单因素实验结合Box-Behnken响应面优化DF的最佳提取工艺,并比较可溶性膳食纤维纯化前(SDF-R)、后(SDF-P)的结构特征、理化性质和功能活性。结果表明:在料液比1∶39(g/mL)、碱液质量分数6%、浸提时间92 min、温度47 ℃的最优工艺下SDF提取率为66.42%。FT-IR表征SDF-R和SDF-P均存在典型的纤维素多糖特征吸收峰结构,未发生主要官能团的构型差异。理化性质方面,SDF-P的持水力、持油力和膨胀力分别较SDF-R提升了2.1%,11.3%和36.7%。抗氧化活性测定表明纯化后SDF-P抗氧化能力下降,DPPH、·OH清除率及铁还原力分别降低了8.6%,13.6%和32.8%,而ABTS+清除率(99%)无显著差异。同样,降血糖功能研究表明SDF-P对α-淀粉酶和α-葡萄糖苷酶抑制率分别降低了4.7%和8.5%,SDF-R表现出较强的抑制活性。结论:玫瑰粗SDF具有优异的功能性和抗氧化特性,可作为理想的膳食纤维应用于功能性食品。

    Abstract:

    In this study, rose petals were used as the raw material to extract dietary fiber(DF) via the chemical alkali method. The optimal extraction process for DF was determined through a combination of single-factor experiments and the Box-Behnken response surface methodology. A systematic comparison of the structural characteristics, physicochemical properties, and functional activities of soluble dietary fiber both pre-purification (SDF-R) and post-purification (SDF-P) was conducted. The results showed that the extraction yield of 66.42% for SDF was achieved under the optimal conditions of a solid-liquid ratio 1∶39(g/mL), alkali concentration 6%, extraction time 92 min, and temperature 47 ℃. FT-IR analysis revealed that both SDF-R and SDF-P exhibited characteristic absorption peaks typical of cellulosic polysaccharides, indicating no significant conformational differences in the primary functional groups. Physicochemical property analysis indicated that the water-holding capacity, oil-holding capacity, and swelling capacity of the SDF-P were increased by 2.1%, 11.3%, and 36.7%, respectively. Antioxidant activity assays revealed a decrease in the antioxidant capacity of SDF-P, with reductions of 8.6%, 13.6%, and 32.8% in the DPPH,·OH scavenging rate, and ferric reducing ability, respectively. In contrast, the ABTS+ scavenging rate(99%) showed no significant difference. Similarly, the hypoglycemic function results indicated that the inhibitory effects of purified soluble dietary fiber SDF-P on α-amylase and α-glucosidase were reduced by 4.7% and 8.5%, respectively, compared to the SDF-R, which exhibited stronger inhibitory activity. In summary, the crude SDF from roses possesses superior functionality and antioxidant characteristics, making it an ideal candidate for application as a dietary fiber in the functional food industry.
