    本文通过湿法美拉德反应对酪蛋白(CN)与低聚半乳糖(GOS)、低聚果糖(FOS)和壳寡糖(COS)进行糖基化处理,比较3种功能性低聚糖对酪蛋白结构及功能特性的影响,筛选出对酪蛋白改性效果最佳的功能性低聚糖。首先测定糖基化产物的接枝率和褐变程度,随后通过扫描电镜、聚丙烯酰氨凝胶电泳(SDS-PAGE)、纳米粒度仪、傅里叶红外光谱和紫外光谱对糖基化产物微观结构进行表征,最后对糖基化产物的功能特性进行了评价。结果表明:CN-FOS接枝率为(29.9±0.65)%,CN-GOS接枝率为(33.5±0.76)%,CN-COS接枝率为(34.2±0.27)%。接枝率的增加导致酪蛋白的性质变化显著,其中CN-COS的变化尤为突出,粒径减小约344 nm;在pH值3~11范围,溶解度均提高,pH值为11时最高为(94.82±0.93);同时其起泡性为226%,提高了58.3%、乳化性为20.18,提高了1.26倍。CN-COS的糖基化产物对DPPH、ABTS和·OH自由基的清除率为64.72%,65.95%,32.38%,分别提高了3.35倍,1.74倍,2.48倍。由结果可知COS对CN的改性效果最好。本研究为功能性低聚糖改性酪蛋白并将其应用于食品行业提供了理论参考。

    Abstract:

    In this study, casein (CN) was glycosylated with galactose oligosaccharide (GOS), fructooligosaccharide (FOS) and chito-oligosaccharide (COS) by wet Maillard reaction. Comparing the effects of three functional oligosaccharides on the structure and functional properties of casein, the most effective functional oligosaccharide for casein modification was selected. First, the grafting rate and browning degree of the glycosylated products were determined. Subsequently, the microstructure of the glycosylated products was characterized using scanning electron microscopy (SEM), sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), a nanoparticle size analyzer, Fourier transform infrared spectroscopy (FTIR), and ultraviolet spectroscopy. Finally, the functional properties of the glycosylated products were evaluated. The experimental results show that the grafting degree of CN-FOS was (29.9±0.65)%, CN-GOS was (33.5±0.76)%, and CN-COS was (34.2±0.27)%. With the increase of grafting degree, the properties of casein change significantly, especially the change of CN-COS, and the particle size decreases by about 344 nm. The solubility increases in a wide pH range, the highest value at pH 11 was (94.82±0.93). Meanwhile, its foaming activity was 226%, an increase of 58.3%, and emulsification activity was 20.18, an increase of 1.26 times. The sugar-glycosylated products of CN-COS showed 64.72%, 65.95%, and 32.38% scavenging rates against DPPH, ABTS, and ·OH free radicals, which increased by 3.35 times, 1.74 times, and 2.48 times respectively. The results indicate that COS demonstrates the most effective modification of CN. This study provides theoretical reference for the modification of casein by functional oligosaccharides and its application in food industry.
