小麦谷氨酰胺类肽的稳定性、肽段结构及抗抑郁活性分子对接研究

    摘要:

    为了扩大小麦谷氨酰胺类肽的应用范围,以谷朊粉为原料制备小麦谷氨酰胺类肽,并研究其在不同温度、酸碱度、金属离子和体外模拟消化条件下的稳定性。通过三重四极杆液相色谱-串联质谱(LC-MS)鉴定小麦谷氨酰胺类肽段,通过分子对接技术研究小麦谷氨酰胺类肽段对抗抑郁活性靶蛋白脑源性神经营养因子(BDNF)的结合位点和结合能力,进而分析小麦谷氨酰胺类肽段的抗抑郁活性。结果表明,小麦谷氨酰胺类肽具有良好的热稳定性、酸碱稳定性和体外消化稳定性,金属Cu2+对其稳定性影响较大。鉴定出Leu-Gly-Gln(LGQ)、PyroGlu-Leu-Gln(pELQ)、PyroGlu-Gln-Leu(pEQL)、PyroGlu-Gln-Gln(pEQQ)、Val-Ala-Gln(VAQ)5条小麦谷氨酰胺类肽,它们与BDNF对接的结合能分别为-5.03,-6.13,-6.55,-6.03,-5.18 kcal/mol,均与BDNF形成稳定的结合物,并通过范德华力,π-烷基等疏水作用力与配体结合,说明小麦谷氨酰胺类肽的5条肽段均具有抗抑郁活性。本研究为小麦谷氨酰胺类肽的生产加工及抗抑郁活性营养健康产品的开发提供理论参考。

    Abstract:

    In order to expand the application range of wheat glutamine-derived peptides, the study prepared wheat glutamine-derived peptides from gluten powder and investigated the temperature, pH, metal ions, and in vitro simulated digestion stability. Wheat glutamine-derived peptide segments were identified using triple quadrupole liquid chromatography tandem mass spectrometry (LC-MS), and the binding sites and binding capacity of the wheat glutamine-derived peptide segments to the antidepressant target protein brain-derived neurotrophic factor (BDNF) were studied using molecular docking technology, further analyzing the antidepressant activity of the wheat glutamine-derived peptide segments. The results showed that wheat glutamine-derived peptide had good thermal stability, acid-base stability and in vitro digestion stability, and metal Cu2+ had a great influence on its stability. Five wheat glutamine-derived peptides, Leu-Gly-Gln (LGQ), PyroGlu-Leu-Gln (pELQ), PyroGlu-Gln-Leu (pEQL), PyroGlu-Gln-Gln (pEQQ) and Val-Ala-Gln (VAQ), were identified, and the binding energies of their binding energies to BDNF were -5.03, -6.13, -6.55, -6.03 and -5.18 kcal/mol, respectively. All of them could form stable binders with BDNF and bind to ligands through van der Waals forces, Pi-Alkyl groups and other hydrophobic forces, indicating that these five peptides of wheat glutamine-derived peptide had antidepressant activity. This study provides theoretical support for the production and processing of wheat glutamine-derived peptides and the development of antidepressant nutritional health products.
