枸杞叶浆水的自然发酵性能与微生物多样性

    摘要:

    为探究枸杞叶浆水自然发酵过程中的发酵特性以及发酵成熟后的微生物结构多样性,测定发酵过程中的各项理化指标、抑菌作用、抗氧化能力,并采用Illumina Miseq技术分析枸杞叶浆水发酵结束后的微生物菌群结构。结果表明,在自然发酵过程中,pH值从4.6降至3.11,总酸含量由0.6 g/L显著提升至5.7 g/L,这一变化表明发酵过程中酸性物质呈持续积累的趋势。与此同时,活菌数量从发酵起始时的1.14×106 CFU/mL增至2.23×106 CFU/mL。抗氧化方面,枸杞叶浆水的DPPH自由基清除率由25.4%升至59%,羟自由基清除率由14.92%增至35.5%,在发酵第6天羟自由基清除率达到峰值(43.34%),还原力从发酵初始的0.22升至第7天的1.177,说明枸杞叶浆水在发酵过程中抗氧化性能力显著增强。微生物群落结构分析显示,枸杞叶浆水自然发酵结束后的绝对优势细菌门是厚壁菌门,相对丰度为99.9%,优势细菌属为乳酸杆菌属,相对丰度为85.65%,粘液乳杆菌属相对丰度为13.46%;子囊菌门占据绝对优势地位,相对丰度为98%,毕赤酵母属相对丰度为87.17%,曲霉属相对丰度为28.63%,丝孢酵母属相对丰度为4.01%,两者为优势真菌属。本研究初步揭示枸杞叶浆水自然发酵过程中的发酵特性演变规律以及发酵结束后的微生物群落结构特征,为丰富浆水资源以及后续筛选具有潜在价值的益生菌菌株提供一定的理论参考。

    Abstract:

    In order to study the fermentation characteristics during the natural fermentation of Lycium barbarum leaf Jiangshui and the microbial diversity at the end of fermentation, the patterns of change of physicochemical indexes, bacteriostatic effect and antioxidant capacity were analyzed and the microbial community structure of Lycium barbarum leaf Jiangshui at the end of fermentation was analyzed using Illumina Miseq high-throughput sequencing technology. The results showed that after 7 days of natural fermentation, the pH decreased from 4.6 to 3.11, the total acid increased from 0.6 g/L to 5.7 g/L, and the number of viable bacteria increased by 2.23×106 CFU/mL from 1.14×106 CFU/mL on the fermentation; in terms of antioxidant, the scavenging rate of the DPPH radical in the Lycium barbarum leaf Jiangshui increased from 25.4% to 59%, and the scavenging rate of hydroxyl radicals increased from 14.92% to 35.5%, the hydroxyl radical scavenging rate reached the highest at 43.34% on the 6th day of fermentation, and the reducing power increased from 0.22 at the beginning of fermentation to 1.177 on the 7th day. The absolute dominant bacterial phylum of Lycium barbarum leaf Jiangshui at the end of the natural fermentation was Bacillota, and the dominant bacterial genus was Lactobacillus, with a relative abundance of 85.65%, and Limosilactobacillus, with a relative abundance of 13.46%; the absolute dominant fungal phylum was Ascomycota, with a relative abundance of 98%, and the dominant fungal genera were Pichia, with a relative abundance of 87.17%, Aspergillus with a relative abundance of 28.63%, and Trichosporon with a relative abundance of 4.01%. This study preliminarily revealed the fermentation characteristics of Lycium barbarum leaf Jiangshui during the natural fermentation process and the microbial community structure at the end of the fermentation, which provided a certain theoretical basis for the enrichment of Jiangshui resources and the subsequent screening of potentially valuable probiotics.
