The Protective Effect and Mechanisms of Hawthorn Flavonoids on DNA Damage of BRL-3A Hepatocytes

    Abstract:

    Objective: To study the protective effect and mechanisms of hawthorn flavonoids on DNA damage of BRL-3A hepatocytes. Methods: MTT method was used to detect the effect of different concentration hawthorn flavonoids on hepatocytes injured by ethanol. Single-cell gel electrophoresis was used to observe the effect of hawthorn flavonoids on the DNA damage of hepatocytes and the Olive tail moment was measured. Physiological and biochemical indexes such as superoxide dismutase (SOD), malonaldehyde (MDA) and glutathione peroxidase (GSH-Px) in cells were detected by kits. The mRNA expression of the gene SOD was measured by real-time quantitative polymerase chain reaction(qPCR). Results: 0.2 mg/mL, 0.4 mg/mL and 0.8 mg/mL hawthorn flavonoids could significantly repair the growth and DNA damage of ethanol induced hepatocytes, which was closely related to the improvement of SOD, MDA and GSH-Px. Conclusion: Hawthorn flavonoids can be used to repair liver cells damaged by ethanol, and hawthorn flavonoids have potential for the treatment of liver injury.
