Determination of 7 Nonpolar Heterocyclic Aromatic Amines in Cantonese Roast Goose by HPLC-Q-Orbitrap-HRMS

    Abstract:

    It is of significance to detect and monitor the heterocyclic aromatic amines (HAAs) contents in Cantonese roast goose because they are very easy to be formed in the high-temperature processing. In order to develop a method on the basis of high-performance liquid chromatography coupled to quadrupole orbitrap high-resolution mass spectrometry (HPLC-Q-Orbitrap HRMS) for the determination of 7 HAAs in Cantonese roast goose, one of the traditional Chinese food, samples were homogenized with NaOH-acetonitrile solution, which could achieve saponification hydrolysis of oil in the samples and extraction of target HAAs. The extraction solution was purified by StrataTM-X-C solid phase extraction cartridge, separated on a C18 column by gradient elution with mobile phase consisting of acetonitrile and 1 mmol/L ammonium acetate containing 0.05% formic acid, detected by electrospray positive ion, full scan and data-dependent scan modes. Retention time, accuracy of primary ion mass in combination with matching of secondary fragments to the library was used for qualitative analysis, and the internal standard method combined with the peak area of primary ion was used for quantitative analysis of HAAs. Results showed that the mass accuracy of the target HAAs was less than 3×10-6, with good linearity in the range of 0.100-100 μg/L, and the correlation coefficients were greater than 0.997. The limits of detection and limits of quantification were 0.02-0.15 μg/kg and 0.05-0.5 μg/kg, respectively. Recoveries of 7 HAAs were good at three spiked concentrations. The established HPLC-Q-Orbitrap HRMS method has high sensitivity, accurate and reliable results, and can be applied to detecting nonpolar HAAs in Cantonese roast goose.
