Screening of Functional Lactic Acid Bacteria for the Production of Pyridoxal Phosphate and Its Probiotic Characteristics

    Abstract:

    Objective:To screen a high pyridoxal phosphate-producing functional lactic acid bacterium and investigate its probiotic characteristics. Methods: The contents of pyridoxal phosphate in 36 lactic acid bacteria stored in our laboratory were determined by high performance liquid chromatography to screen a high pyridoxal phosphate-producing functional lactic acid bacterium, and its probiotic characteristics were then studied. Results: A lactic acid bacterium N11 with a high yield of pyridoxal phosphate of 313.52 mg/L was screened. The one-step growth curve indicated that the growth of the strain N11 was the lag phase at the first 6 h, and then entered the log growth phase after 6 h and the stationery phase after 18 h. The optimal growth temperature and pH were 37 ℃ and 6.0, respectively. The self-coagulation rate at the culture of 24 h was up to 41.13%. The tolerant mass fraction of the strain N11 to hydrogen peroxide, NaCl, pH and bile salt were 1.0 mmol/L, 8%, 3.0 and 0.1%, respectively. After frozen at -20 ℃ for 5 d, the number of viable bacteria was still 106. This strain could obviously inhibit the growth of Pseudomonas fluorescens, Escherichia coli, Listeria monocytogenes, Staphylococcus aureus and Salmonella, with a diameter of the inhibitory zone of more than 15 mm. Conclusion: These findings lay good foundation for the development and application of functional lactic acid bacteria that can highly produce pyridoxal phosphate.
