Abstract:In order to select fine Lactobacillus with local characteristics, Shunde buffalo milk and Jieyang goat milk were selected as samples in this study. After 16S rRNA identification, 3 strains of Lactobacillus salivarius, 3 strains of Lactobacillus fermentum, 3 strains of Lactobacillus rhamnosus, 3 strains of Lactobacillus plantarum and 37 strains of Lactobacillus paracasei were screened from Shunde buffalo milk. In Jieyang goat milk, 8 strains of Lactobacillus fermentum, 11 strains of Lactobacillus paracasei and 5 strains of Lactobacillus plantarum were screened. These Lactobacillus from two sources were selected for heat resistance, acid resistance, bile salt resistance, hemolytic resistance and drug sensitivity tests. The results showed that Lactobacillus paracasei R21 and JY8 from buffalo milk and goat milk had good heat resistance, and the survival rate remained above 100% after heating at 50 ℃ for 20 min. The survival rate of 8 strains of Lactobacillus was above 77% after 2 h culture with pH 2.5, and the survival rate of Lactobacillus salivarius S54 was 112.28%. After being treated with 0.1% bile salt for 2 h, the bile salt tolerance of Lactobacillus salivarius S54 and Lactobacillus paracasei JY8 was poor, but the bile salt tolerance of other strains was good. The Lactobacillus derived from the two samples have good safety, no hemolytic circles are produced on the Columbia blood plate, and they are sensitive to multiple antibiotics. It can be seen that there are abundant lactic acid bacteria resources in buffalo milk and goat milk from Guangdong province, and some strains have good heat and acid resistance, which can be used as outstangding probitic and fermentation starter germplasm resources.
